SONATA grant for KETO-MINOX 2.0

The ketogenic diet (KD) has recently gained substantial popularity as a weight‑management strategy, yet its metabolic and physiological effects remain insufficiently characterized. Most existing studies investigate extremely low‑calorie ketogenic regimens, making it difficult to distinguish the impact of ketosis from the consequences of severe caloric restriction. This project, continuing the KETO‑MINOX study, aims to provide a comprehensive evaluation of an eight‑week, isocaloric, energy‑restricted KD in women with overweight and obesity, focusing on metabolic outcomes and gut microbiota composition.

The study was conducted as a randomized, controlled nutritional intervention comparing a 1750 kcal KD (F:P:C ratio 70:20:10) with a standard balanced diet (20:30:50). Biological samples were collected at baseline (T0), after four weeks (T1), after eight weeks (T2), and at a one‑year follow‑up (T3). 

The project will apply a multiomics approach, including non‑targeted metabolomics, lipidomics, volatilomics, targeted fatty acid analysis, microbiome sequencing, and chemometric integration. By leveraging previously collected samples, the project will generate robust, long‑term insights into the safety and metabolic consequences of KD, contributing valuable evidence to nutrition science and obesity research.
